Overview

Pru p 3-specific B-cell receptors (BCRs) are membrane-bound immunoglobulins on B lymphocytes that specifically recognize Pru p 3, the primary non-specific lipid transfer protein (nsLTP) allergen in peach (Prunus persica) (UniProt P81402). These receptors are central to the allergic sensitization process, where they capture the Pru p 3 allergen and initiate B-cell signaling pathways leading to the production of allergen-specific IgE (PMID: 24565318). In sensitized individuals, the interaction between Pru p 3 and these BCRs triggers a cascade of immune responses that can result in symptoms ranging from oral allergy syndrome to life-threatening anaphylaxis (PMID: 28610661). In clinical settings, these BCRs are the primary targets for allergen-specific immunotherapy (AIT), which seeks to reprogram the immune system toward tolerance by shifting the response from IgE production to protective IgG4 production (PMID: 25172335). Successful AIT leads to a reduction in IgE-producing B cells and an increase in regulatory B cells (Bregs) specific to Pru p 3. Emerging therapies also include the use of Pru p 3-derived peptides to target these receptors without cross-linking IgE on mast cells, thereby reducing the risk of systemic reactions (PMID: 21920024). Understanding the repertoire and affinity of these BCRs is essential for developing precision treatments for peach-induced allergic diseases.
