Pseudomonas aeruginosa type IV pili (T4P) are dynamic, proteinaceous surface appendages that play a fundamental role in the virulence and environmental adaptation of this Gram-negative pathogen (Burrows, 2012, Annu Rev Microbiol). These structures are primarily composed of the major pilin subunit, PilA, and are assembled through a complex machinery that enables the extension and retraction of the pilus fiber (Craig et al., 2019, Nat Rev Microbiol).\n\nThe primary biological function of T4P is twitching motility, a unique form of surface translocation powered by the rapid retraction of the pili, which is essential for the initial stages of host colonization and the formation of robust biofilms (O'Toole and Kolter, 1998, Mol Microbiol). Additionally, T4P serve as critical adhesins that mediate the attachment of the bacteria to host epithelial cells by binding to specific glycolipid receptors like asialo-GM1 (Luo et al., 2015, Infect Immun). Given their necessity for infection, T4P are considered high-priority targets for anti-virulence therapies, including monoclonal antibodies and small-molecule inhibitors aimed at disrupting pilus assembly or function (Kuo et al., 2020, J Biol Chem). Such therapeutic approaches aim to attenuate the pathogen's ability to cause disease and evade the immune system, potentially providing an alternative or adjunct to traditional antibiotics (Baron, 2010, Future Microbiol).
