The purine transporter system in trypanosomes refers to a group of membrane proteins responsible for the uptake of purine nucleobases and nucleosides from the host environment. Trypanosomatid parasites, such as *Trypanosoma brucei*, lack the ability to synthesize purines de novo and are entirely dependent on salvaging these essential molecules from their hosts. This makes their purine transporters critical for parasite survival, proliferation, and infectivity. The best-characterized systems in *T. brucei* are the P1 transporter system (broad specificity), the P2 transporter system (TbAT1; high affinity for adenosine and certain drugs), and nucleobase transporters such as NT8 and NT10. These transporters are essential for acquiring purines, signaling, and drug uptake, making them promising therapeutic targets. Drug resistance can arise from mutations or downregulation of these transporters.
