Overview

PVR related immunoglobulin domain-containing protein (PVRIG), also known as CD112R, is a novel immune checkpoint receptor belonging to the poliovirus receptor (PVR)-like family. It is primarily expressed on T cells and natural killer (NK) cells, where it interacts with its ligand, PVRL2 (CD112), to inhibit immune cell activation and effector functions. In the tumor microenvironment, the PVRIG-PVRL2 pathway serves as a mechanism for tumor immune evasion, often acting in a complementary fashion to the TIGIT-PVR axis. Historically, PVRIG was referred to as an "undisclosed first-in-class immune cell surface antigen" by companies like Compugen during its early discovery and development phases [1]. Therapeutic strategies targeting PVRIG, such as the monoclonal antibody COM701, aim to block this inhibitory signal and restore anti-tumor immunity, particularly in patients who are non-responsive to standard PD-1/PD-L1 inhibitors [2].
