Pvs230 protein (Plasmodium vivax) (Pvs230)

Target
Pvs230
Molecular classification
Gametocyte/gamete surface antigen, 6-cysteine protein family, Transmission-blocking vaccine target
01

Overview

Pvs230 is a large, multidomain surface protein expressed by Plasmodium vivax on gametocytes and gametes, belonging to the 6-cysteine (6-Cys) protein family, characterized by multiple conserved cysteine-rich domains[1][4]. Its N-terminal domain (domain 1, or Pvs230D1) contains immunogenic epitopes that elicit transmission-blocking antibodies and is under study as a major candidate antigen for malaria transmission-blocking vaccines[2][4]. The biological function mirrors its Plasmodium falciparum ortholog (Pfs230), being essential for male gamete fertility, gamete recognition, and possibly parasite binding to human erythrocytes. Antibodies against Pvs230, particularly against the D1 domain, can block parasite transmission to mosquitoes, making it a leading candidate for next-generation malaria vaccines aimed at interrupting transmission[2][4].

02

Mechanism of action

Transmission blocking via antibody-mediated inhibition of parasite fertilization in the mosquito

03

Biological functions

Surface localization on sexual-stage parasites (gametocytes and gametes)Involved in parasite fertilization and transmission to mosquitoesPotential role in gamete recognition and host cell (erythrocyte) interaction
04

Disease associations

Infection (malaria; specifically Plasmodium vivax transmission)
05

Safety considerations

No major safety concerns documented for targeting Pvs230; potential challenge is antigenic diversity and the need to induce functional, complement-fixing antibodies
06

Interacting drugs

None currently approved; monoclonal antibodies (experimental) targeting this protein
07

Biomarkers

None established for clinical use; anti-Pvs230 antibody levels explored as surrogate of transmission-blocking immunity
