Overview

PXN antisense RNA 1 is a long non-coding RNA located on chromosome 12 (hg38: chr12:120201260-120213401), transcribed opposite to the Paxillin (PXN) gene. It undergoes alternative splicing regulated by MBNL3, resulting in at least two major transcript isoforms (PXN-AS1-L and PXN-AS1-S). PXN-AS1-L upregulates PXN protein levels by stabilizing PXN mRNA, particularly via direct binding to the 3’UTR and protection against miRNA-induced degradation. PXN-AS1-S can decrease PXN mRNA translation. Together, their interplay helps maintain PXN homeostasis in adult liver tissue, and their dysregulation is implicated in the progression of hepatocellular carcinoma and other cancers. PXN-AS1 may serve as a biomarker for poor prognosis in cancer patients and as a putative therapeutic target due to its critical role in cancer biology.
