Pyridoxal 5’-phosphate-dependent enzymes are a large and diverse family of enzymes that require pyridoxal 5'-phosphate (PLP), the active form of vitamin B6, as a coenzyme. PLP is essential for catalyzing a wide range of biochemical reactions, particularly those involving amino acids. These enzymes play central roles in amino acid metabolism, biosynthesis of neurotransmitters, synthesis and catabolism of sphingolipids and heme. The key feature is the formation of a Schiff base (aldimine) between the aldehyde group on PLP and an active site lysine residue on the enzyme. PLP acts as an electron sink to stabilize carbanionic intermediates formed during various reaction types. Deficiency or dysfunction in these enzymes can lead to metabolic disorders due to impaired neurotransmitter synthesis or amino acid metabolism.
