Quaking (QKI) is a KH domain-containing RNA-binding protein and member of the STAR family, with key roles in regulating RNA metabolism, including pre-mRNA splicing, stability, export, and translation. It is essential for normal development of the nervous system—particularly for oligodendrocyte differentiation and myelination—by controlling RNA homeostasis at transcriptional and post-transcriptional levels. QKI functions as a tumor suppressor and regulates cellular processes such as angiogenesis, apoptosis, and cell differentiation. QKI interacts extensively with microRNAs, long non-coding RNAs, and circular RNAs, forming intricate regulatory networks relevant in cancer, neurological, and developmental diseases. The MYB-QKI fusion protein is specifically implicated in certain gliomas. QKI is emerging as a valuable biomarker for diagnosing and monitoring several cancers and CNS disorders[1][2].
