Raftlin, lipid raft linker 1 (RFTN1) is a membrane-associated scaffold protein predominantly found in immune cells, especially B and T lymphocytes. It plays a key role in the formation and stability of lipid rafts—cholesterol-rich membrane microdomains that serve as platforms for cell signaling. In B cells, Raftlin is critical for the proper assembly of lipid rafts, which in turn regulate antigen-dependent B-cell receptor (BCR) signaling and cell proliferation. Its localization is determined by N-terminal acylation (myristoylation and palmitoylation). Raftlin participates in the trafficking and internalization of Toll-like receptors and double-stranded RNA, thereby linking membrane microdomains to innate immune signaling. Genetic association studies have implicated it as a biomarker for glaucoma, with other studies noting its association with some hereditary red blood cell disorders and chronic otitis media. No drugs are known to interact directly with Raftlin, and it is not considered a classical drug target such as an enzyme, receptor, or transporter.
