Overview

RalA-binding protein 1 (RALBP1), commonly referred to as RLIP76, is a multifunctional scaffold protein and a key effector of the Ral signaling pathway (UniProt Q15311). It functions as a non-ABC (ATP-binding cassette) transporter, mediating the ATP-dependent efflux of glutathione-conjugates and various hydrophobic drugs, which contributes to multidrug resistance in malignancies (PubMed: 25100370). RALBP1 also possesses GTPase-activating protein (GAP) activity towards Rac1 and Cdc42, thereby regulating cytoskeletal dynamics and clathrin-dependent endocytosis (PubMed: 15632142). In many cancers, including lung, colon, and prostate cancer, RALBP1 is overexpressed and promotes cell survival, proliferation, and metastasis (PubMed: 21810915). Therapeutic strategies targeting RALBP1, such as antisense oligonucleotides or specific antibodies, have demonstrated the ability to induce apoptosis and enhance the efficacy of radiation and chemotherapy in preclinical studies (PubMed: 19448679). Its role in metabolic regulation also suggests potential implications in diabetes and oxidative stress-related disorders (PubMed: 23603127). Given its diverse roles in cellular detoxification and signaling, RALBP1 represents a promising therapeutic target for overcoming drug resistance in oncology.
