Overview

Ran-binding protein 17 (RANBP17) is a member of the importin-beta superfamily of nuclear transport receptors, which facilitate the movement of proteins across the nuclear pore complex (UniProt Q9H2S4). It is characterized by its high expression in the testis, indicating a specialized role in male germ cell development and spermatogenesis (PubMed: 11053416). In the context of human disease, RANBP17 is frequently associated with B-cell precursor acute lymphoblastic leukemia (BCP-ALL) through the IGH-RANBP17 translocation, which often results in the overexpression of the CRLF2 gene (PubMed: 19933319). This genetic rearrangement places the RANBP17 locus under the control of the immunoglobulin heavy chain enhancer, contributing to leukemogenesis (PubMed: 20008148). Although RANBP17 is not currently a direct target for any approved therapeutic agents, its involvement in oncogenic signaling pathways and nuclear transport makes it a potential candidate for future research into targeted inhibitors. Therapeutic challenges include the high sequence homology among importin-beta family members, which may lead to off-target effects if specificity is not achieved.
