Overview

The Ras homolog family member G (RHOG) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the transcript encoding the RhoG protein, a small GTPase of the Rho family. This region contains evolutionary conserved binding sites for various microRNAs, most notably miR-124, which post-transcriptionally repress RhoG expression by promoting mRNA degradation or inhibiting translation (NCBI Gene ID: 391; PubMed: 24510035). RhoG itself plays a pivotal role in cytoskeletal remodeling, lamellipodia formation, and cell migration by acting upstream of the Rac1 signaling pathway. In several malignancies, such as glioblastoma and breast cancer, the downregulation of regulatory microRNAs leads to the overexpression of RhoG via its 3'-UTR, thereby driving tumor cell invasion and metastasis (PubMed: 25110378). Consequently, the RHOG mRNA 3'-UTR is considered a therapeutic target for RNA-based interventions, including microRNA mimics and antisense oligonucleotides, designed to restore normal RhoG levels and suppress oncogenic phenotypes.
