Overview

RAS p21 protein activator 3 (RASA3) is a member of the GAP1 family of GTPase-activating proteins that serves as a critical negative regulator of Ras and Rap1 signaling [1, 2]. By facilitating the transition of these small GTPases from their active GTP-bound state to their inactive GDP-bound state, RASA3 modulates essential cellular processes including adhesion, migration, and proliferation [1, 3]. In the context of hematology, RASA3 is vital for maintaining platelet quiescence; its loss or downregulation leads to constitutive Rap1 activation, resulting in platelet hypersensitivity and increased thrombotic risk [2]. Furthermore, RASA3 has been implicated in vascular development and various cancers, where it often functions as a tumor suppressor whose expression is frequently reduced to promote oncogenic signaling [4]. While no small molecules currently target RASA3, its mRNA represents a viable therapeutic target for RNA-based interventions, such as antisense oligonucleotides or mRNA replacement therapies, aimed at restoring or modulating its regulatory function in cardiovascular and oncological diseases [2, 4].
