Overview

Ras-related protein Rab-38 (RAB38) is a small GTPase belonging to the Ras superfamily that serves as a master regulator of intracellular membrane trafficking, particularly in melanocytes and lung alveolar type II cells (UniProt Consortium, 2023). It is essential for the sorting and transport of melanogenic enzymes, such as tyrosinase and tyrosinase-related protein 1 (TYRP1), from the trans-Golgi network to melanosomes, thereby playing a vital role in pigmentation (Wasmeier et al., 2006). Mutations in the RAB38 gene are linked to Hermansky-Pudlak syndrome-like phenotypes, characterized by oculocutaneous albinism and platelet storage pool deficiency (Osanai et al., 2010). In the context of oncology, RAB38 is identified as the tumor-associated antigen NY-MEL-1, which is highly expressed in melanoma but has limited expression in normal tissues (Jäger et al., 2000). This restricted expression profile makes it an attractive target for immunotherapy, including cancer vaccines and T-cell therapies designed to elicit an immune response against malignant cells (Gnjatic et al., 2003). Research into RAB38-targeted therapies focuses on leveraging its role as a source antigen to induce specific anti-tumor immunity while monitoring for potential autoimmune effects like vitiligo.
