Overview

The RAB7B 13–21 peptide presented by HLA-A*11:01 is a peptide-major histocompatibility complex (pMHC) that serves as a critical self-antigen in the development of T-cell receptor (TCR) therapies (Shen et al., 2025, J Immunother Cancer). It consists of a 9-mer peptide (VIVGAIGVG) derived from the Ras-related protein Rab-7b, which is endogenously processed and presented on the surface of cells expressing the HLA-A*11:01 allele (UniProt Q96AH8). This complex is particularly notable for its high structural and sequence homology to the KRAS G12V neoantigen (VVVGAVGVG), a common driver mutation in solid tumors (Bear et al., 2024, JCI). Because of this similarity, TCRs engineered to target KRAS G12V often exhibit off-target cross-reactivity with the RAB7B complex. Since RAB7B is expressed in healthy tissues, including dendritic cells and monocytes, such cross-reactivity can lead to severe immune-mediated toxicities. Therefore, the RAB7B 13–21/HLA-A*11:01 complex is primarily used as a safety screening target to identify and exclude cross-reactive TCR candidates during the preclinical development of KRAS-targeted immunotherapies.
