Overview

The Ras signaling pathway is a central cellular signaling cascade that regulates cell proliferation, survival, differentiation, apoptosis, cytoskeletal dynamics, and gene expression. It is activated by growth factors and other extracellular signals leading to GTP-binding and activation of Ras proteins (KRAS, NRAS, HRAS). Activated Ras then triggers downstream pathways such as MAPK/ERK, PI3K/AKT/mTOR, and others. Dysregulation of this pathway, often due to mutations in Ras genes, is implicated in cancer and developmental disorders. Therapies targeting components of this pathway are used in cancer treatment, but resistance and safety concerns remain significant challenges.
