Overview

Recepteur d'Origine Nantais (RON) protein tyrosine kinase, also known as MST1R, is a receptor tyrosine kinase (RTK) with significant roles in cell signaling, cancer progression, and metastasis. It regulates inflammation, cell growth, migration, epithelial-to-mesenchymal transition (EMT), tissue repair, and immune responses. RON is synthesized as a single-chain precursor, which is cleaved into an extracellular α-chain and a transmembrane β-chain containing the intracellular tyrosine kinase domain. Binding of Macrophage-stimulating protein (MSP) to RON leads to receptor dimerization and autophosphorylation on key tyrosines in the cytoplasmic domain, activating downstream pathways such as MAPK/ERK and PI3K/AKT. RON overexpression/activation drives oncogenic processes, making it a target for anti-cancer therapies.
