Overview

The receptor-binding domain (RBD) of the SARS-CoV-2 spike protein is a critical region responsible for binding to the host cell receptor, angiotensin-converting enzyme 2 (ACE2), facilitating viral entry into human cells. The Beta variant, also known as lineage B.1.351 or 501Y.V2, was first identified in South Africa and is classified as a Variant of Concern due to its increased infectivity and ability to evade immune responses. The RBD encompasses amino acids 319–541 of the spike glycoprotein and contains three key mutations: K417N, E484K, and N501Y. These mutations alter the structure and stability of the RBD, affecting both ACE2 binding affinity and antibody recognition, leading to enhanced infectivity and reduced neutralization by some antibodies.
