Regenerating family member 1 gamma, pseudogene (REG1CP) is a transcribed long non-coding RNA (lncRNA) located within the human REG gene cluster. Formerly considered a pseudogene, REG1CP is now recognized as having a functional regulatory role, particularly in the promotion of cancer cell proliferation and tumorigenesis. Mechanistically, REG1CP forms an RNA–DNA triplex with the promoter of the REG3A gene, recruiting FANCJ helicase to unwind DNA and facilitate glucocorticoid receptor α-mediated transcription of REG3A. This activity selectively increases REG3A expression, resulting in enhanced cell proliferation, and is implicated in poor prognosis in colorectal cancer. REG1CP is not a traditional therapeutic target (e.g., receptor, enzyme, transporter) but is a non-coding RNA regulator, and its categorization as a pseudogene has changed due to the discovery of its functional transcript[1][2][3][4][5][6].
