Overview

Repulsive guidance molecule b (RGMb), also known as Dragon, is a glycosylphosphatidylinositol (GPI)-anchored membrane protein and a member of the repulsive guidance molecule family [1, 2]. It primarily functions as a co-receptor for bone morphogenetic proteins (BMPs) and neogenin, playing a critical role in nervous system development, axon guidance, and the regulation of cell adhesion [1, 5]. In the immune system, RGMb has been identified as a binding partner for programmed death-ligand 2 (PD-L2), where it contributes to the regulation of respiratory immunity and T-cell tolerance [3]. Due to its involvement in promoting tumor cell proliferation and its role in immune checkpoint pathways, RGMb is being investigated as a therapeutic target in various cancers, such as non-small cell lung cancer, and inflammatory diseases [4]. Therapeutic strategies often involve monoclonal antibodies designed to block its interaction with BMPs or PD-L2 to modulate signaling pathways or enhance anti-tumor immune responses [3, 4]. Additionally, its role in the central nervous system makes it a potential target for treating spinal cord injuries and chronic pain [2, 5].
