Overview

Retinoblastoma-binding protein 6 (RBBP6) is a multi-functional scaffold protein essential for mRNA processing, protein ubiquitination, and cell cycle regulation [UniProt, Q7Z6E9]. It contains a RING finger domain that functions as an E3 ubiquitin ligase, facilitating the degradation of key proteins such as the tumor suppressor p53 [Gao et al., 2013]. RBBP6 is frequently overexpressed in various human malignancies, including breast, colon, and lung cancers, where it promotes cell proliferation and inhibits apoptosis, making it a significant target for oncological intervention [Gao et al., 2013]. Beyond its role in cancer, RBBP6 has been identified as a critical host factor for the Ebola virus, where it interacts with the viral transcription factor VP30 to regulate viral gene expression [Ilinykh et al., 2015]. Therapeutic strategies focusing on RBBP6 mRNA involve the use of small interfering RNAs (siRNAs) or antisense oligonucleotides to silence its expression, thereby inhibiting tumor growth or viral assembly [Gao et al., 2013; Ilinykh et al., 2015]. However, therapeutic development must account for the protein's essential role in development, as RBBP6 deficiency leads to embryonic lethality in animal models [NCBI Gene, 5930].
