Overview

Retinoic acid receptor alpha (RAR-alpha) is a ligand-activated transcription factor belonging to the nuclear receptor superfamily (UniProt: P10276). In keratinocytes, it is a primary RAR isoform and serves as a master regulator of epidermal homeostasis, controlling the balance between cell proliferation and terminal differentiation (PubMed: 8901442). When activated by ligands such as all-trans retinoic acid, RAR-alpha forms heterodimers with retinoid X receptors (RXR) to bind retinoic acid response elements (RAREs) in the promoter regions of target genes (PubChem: CID 5282379). This signaling pathway is essential for maintaining the skin barrier, and its disruption is linked to disorders like psoriasis and ichthyosis (NIH: StatPearls). Therapeutic agents targeting RAR-alpha, including topical and systemic retinoids, are widely used to treat acne and photoaging by promoting keratinocyte turnover and reducing follicular plugging (PubMed: 16281880). Despite their efficacy, RAR-alpha modulators carry significant safety risks, most notably severe teratogenicity and mucocutaneous toxicity (FDA: Retinoid Safety).
