The non-nucleoside site on HIV-1 reverse transcriptase is a specific allosteric, hydrophobic pocket targeted by non-nucleoside reverse transcriptase inhibitors (NNRTIs). NNRTIs bind to this site and induce conformational changes that disrupt the polymerase activity of reverse transcriptase, inhibiting viral DNA synthesis. This site is unique to HIV-1 RT, making NNRTIs selective for HIV-1. NNRTIs do not inhibit human cellular polymerases.
