Overview

Rho-related GTP-binding protein RhoG (RHOG) is a member of the Rho family of small GTPases that plays a pivotal role in regulating the actin cytoskeleton and intracellular signaling (UniProt P84095). It acts as a molecular switch, cycling between an inactive GDP-bound state and an active GTP-bound state to mediate processes such as cell migration, neurite outgrowth, and endocytosis (NCBI Gene ID: 391). RHOG is particularly noted for its ability to activate Rac1 through the ELMO/Dock180 complex, thereby promoting lamellipodia formation and cell motility (PubMed: 11832213). In oncology, RHOG mRNA and protein are often overexpressed, contributing to the invasive and metastatic potential of various tumors, including glioblastoma and breast cancer (PubMed: 23603116). Consequently, RHOG mRNA has emerged as a potential therapeutic target, with research focusing on the use of small interfering RNAs (siRNAs) to silence its expression and inhibit tumor progression (PubMed: 25670011). Despite its therapeutic potential, challenges remain regarding the delivery of RNA-based inhibitors and the potential for off-target effects within the complex Rho GTPase signaling network.
