Ribosomal protein S4, Y-linked 2 (RPS4Y2) is a structural component of the small (40S) ribosomal subunit in human cells, encoded by the RPS4Y2 gene on the Y chromosome[2][3][4]. This protein belongs to the S4E family and is highly similar to its paralogue RPS4Y1; both are Y-linked, male-specific genes, with RPS4Y2 displaying a more restricted expression primarily in the testes and prostate[1][3][4]. RPS4Y2 is believed to play a specialized role in spermatogenesis, possibly impacting germ cell development and male fertility[1][3]. Unlike most ribosomal proteins that are constitutively expressed, RPS4Y2's expression is regulated by alterations in its promoter region, which may have enabled its testis-specific activity[1]. Mutations or haploinsufficiency in ribosomal protein S4 genes have been implicated, albeit controversially, in certain genetic disorders such as Turner syndrome, and more specifically with Y-linked spermatogenic failure[2][3]. There are no known drugs targeting this protein, and it is not currently considered a direct therapeutic target[3][4].
