RNA, 5S ribosomal pseudogene 375 (RNA5SP375) is a non-coding, non-functional genetic sequence classified as a pseudogene within the human genome, derived from the 5S ribosomal RNA gene family[2][6]. Unlike the canonical 5S ribosomal RNA, which is a critical structural component of ribosomes and engages in protein synthesis[1][3][5], RNA5SP375 has lost coding ability and is not transcribed into a functional RNA product. Pseudogenes such as RNA5SP375 typically arise through duplication or retrotransposition events, followed by genetic changes (mutations, truncations, or early stop codons) rendering the sequence inactive and non-essential, with no demonstrated biological or disease role[4][6]. While functional 5S rRNA is highly conserved and plays a pivotal part in ribosome assembly and cellular translation, the pseudogene variant is retained as non-functional genomic DNA and serves as a genetic relic. RNA5SP375 is cataloged in genomic databases with aliases such as RN5S375, but should not be confused with any druggable or disease-relevant target[2][6].
