RNA, 5S ribosomal pseudogene 468 (RNA5SP468) is a human genomic locus that originated from retrotransposition of a 5S ribosomal RNA gene, resulting in a processed pseudogene[1][2][7]. Unlike functional rRNA genes, processed pseudogenes lack promoters and may accumulate sequence deletions or mutations rendering them non-translatable and non-functional[5][6]. Pseudogenes of 5S rRNA, such as RNA5SP468, are common in mammalian genomes and represent a record of the evolutionary dynamics of rRNA gene families, but do not generally contribute to ribosome function or cellular biology[5][6]. These loci are typically not transcribed and have no protein product. While pseudogenes as a group have been implicated in some regulatory functions and disease associations through non-coding RNA activity or as regulators of their wild-type counterparts, no evidence links RNA5SP468 specifically to disease, biomarker utility, or therapeutic applications[8]. 5S ribosomal RNA is a highly conserved component of ribosomes, structurally essential for large subunit function[3][5]. Functional 5S rRNA genes in humans are distinct from their pseudogene counterparts, such as RNA5SP468, which are generally nonfunctional remnants retained in the genome after gene duplication and degeneration[5][6]. Genomic databases such as GeneCards, NCBI Gene, and BioGPS designate RNA5SP468 as a pseudogene, with no associated protein product or therapeutic relevance[1][2][7]. RNA5SP468 does not encode a receptor, enzyme, or transporter and should not be considered a conventional drug target[1][2][7].
