Overview

RNA, U6 small nuclear 661, pseudogene (RNU6-661P) is a DNA sequence in the human genome closely related to the U6 small nuclear RNA (snRNA) gene family, sharing high sequence homology but lacking protein-coding potential and canonical RNA function due to mutations or truncations. U6 snRNA itself is highly conserved, provides core catalytic functions in the spliceosome for intron removal from pre-mRNA, and is transcribed by RNA polymerase III[1][2]. Many vertebrate genomes contain pseudogenes derived from U6 snRNA, such as RNU6-661P, but these serve mainly as evolutionary relics of gene duplication or retrotransposition events and are typically not functionally expressed nor catalytically active[2][3]. There is no evidence that RNU6-661P, as a pseudogene, plays an active role in human disease, drug response, or cellular physiology[3][4]. RNU6-661P is best classified as a non-coding RNA pseudogene and not as a functional therapeutic target, biomarker, or druggable entity. If functional investigation is required, canonical U6 snRNA (not pseudogenes) should be referenced for splicing and molecular function[1][2][4].
