Overview

RNA 5'-phosphate and 3'-OH ligase 1 (RLIG1) is an ATP-dependent enzyme that catalyzes the ligation of RNA strands containing a 5'-phosphate and a 3'-hydroxyl group, via a three-step nucleotidyl transfer mechanism involving an enzyme-AMP intermediate. RLIG1 is selective for single-stranded RNA substrates, does not seal nicks in DNA-RNA hybrids, and may play a role in RNA repair and integrity, particularly during oxidative stress. Genetic evidence and biochemical studies implicate RLIG1 in tRNA metabolism, especially in the brain, with potential, though not well-characterized, links to human disease[1][2][5].
