Overview

RNA polymerase II, I and III subunit K (POLR2K) is one of the smallest core subunits shared by all three major eukaryotic DNA-directed RNA polymerases (I, II, and III), which are essential for synthesizing messenger RNA, ribosomal RNA precursors, and many functional non-coding RNAs[2][4][1][5]. POLR2K does not act as an independent enzyme or receptor but is instead a ubiquitous, indispensable subunit of the multi-protein RNA polymerase complexes required for transcription of nuclear genes in eukaryotic cells. Pathogenic variants may cause severe syndromic disorders, though direct pharmacological targeting is not currently considered tractable due to the essential nature of the subunit[2][4].
