Overview

Rotavirus G1P antigens are the primary outer capsid proteins, VP7 (G-protein) and VP4 (P-protein), of the Rotavirus A species, specifically belonging to the G1 and P (typically P[8]) genotypes. VP7 is a 37-kDa glycoprotein that forms the smooth surface of the virus, while VP4 is an 88-kDa protease-sensitive protein that forms spikes extending from the surface [UniProt P04502, P12473]. These proteins are critical for viral pathogenesis; VP4 mediates attachment to host cell receptors, and VP7 facilitates membrane penetration and uncoating [Crawford et al., 2017]. In the context of human disease, the G1P[8] combination is the most prevalent genotype globally, responsible for a significant burden of severe dehydrating gastroenteritis in infants [Bányai et al., 2012]. These antigens are the principal targets of the host's protective immune response, particularly the production of neutralizing IgA and IgG antibodies that prevent viral entry into enterocytes [Angel et al., 2007]. Consequently, they serve as the basis for major live-attenuated vaccines, such as Rotarix (a monovalent G1P[8] human strain) and RotaTeq (a pentavalent bovine-human reassortant). These vaccines utilize the G1P antigens to induce long-term immunological memory, drastically reducing rotavirus-related hospitalizations and mortality worldwide [Burny et al., 2014].
