Overview

Rotavirus G9 refers to the G9 serotype of the major outer capsid glycoprotein VP7, a 37 kDa protein that plays a critical role in the attachment and entry of Group A Rotaviruses into host cells [1, 11]. It is arranged in a calcium-stabilized T=13 icosahedral trimer that forms the outermost layer of the virion, where it acts as a gatekeeper for the uncoating process [11, 12]. Upon infection, the low calcium environment of the endosome triggers the dissociation of the VP7 trimer, which is a prerequisite for the conformational changes in the VP4 spikes necessary for membrane penetration [11, 15]. As a primary target for the host immune system, the G9 serotype is highly immunogenic and elicits neutralizing antibodies that provide protection against diarrheal disease [7, 8]. The global emergence of G9 as a dominant epidemiological strain has led to its inclusion in modern vaccine formulations, such as the ROTAVAC vaccine derived from the G9P[11] 116E strain [1, 14]. Therapeutic management primarily involves the use of live attenuated vaccines to stimulate mucosal immunity, though research continues into monoclonal antibodies and subunit vaccines that stabilize the VP7 structure to prevent viral infectivity [11, 13].
