Overview
Rotavirus is a nonenveloped, triple-layered virus with a double-stranded RNA genome. The outer capsid comprises two key proteins: VP4, the spike protein, mediates cell attachment and penetration, and functions as a hemagglutinin; VP7, the glycoprotein, forms the outer shell and stabilizes the viral particle through Ca^2+^ ions. Both VP4 and VP7 independently elicit neutralizing antibody responses, forming the basis of serotype-specific immunity and vaccine development. VP4 and VP7 genetic diversity is the foundation for G (VP7) and P (VP4) serotyping of rotavirus strains, guiding both surveillance and vaccination strategies.
