Overview

The VP8* domain is a proteolytically cleaved fragment of the rotavirus outer capsid spike protein, VP4. It forms the globular head of each spike on the virion surface and is essential for viral attachment to host cells. VP8* adopts a galectin-like fold and mediates initial attachment to host cell surface glycans, including histo-blood group antigens (HBGAs) and sialic acids, determining host range and tissue tropism. Interaction between rotavirus's VP8* domain and specific cell-surface glycans is critical for infection initiation, informing vaccine design strategies aimed at blocking viral entry.
