Overview

S100A2, S100A6, and S100P are low-molecular-weight, EF-hand calcium-binding proteins belonging to the S100 family, which are primarily involved in the regulation of the cell cycle, proliferation, and motility [4, 5, 18]. These proteins are frequently overexpressed in various cancers, such as pancreatic, lung, and colorectal carcinomas, where they facilitate tumor growth, invasion, and metastasis by interacting with intracellular targets like p53 or extracellular receptors like the Receptor for Advanced Glycation End Products (RAGE) [1, 3, 6, 7]. S100P is particularly notable for its role in pancreatic cancer progression and its interaction with RAGE, which activates pro-survival and pro-metastatic signaling pathways [12, 13]. S100A6, also known as calcyclin, and S100A2 are also implicated in tumorigenesis and serve as potential diagnostic and prognostic biomarkers in non-small cell lung cancer and other malignancies [10, 11, 14, 16]. Therapeutic strategies targeting these proteins include small molecule inhibitors like cromolyn and V-9, which disrupt protein-protein interactions, and neutralizing antibodies aimed at blocking their extracellular signaling functions [4, 7, 19]. However, the high structural homology among the 20+ S100 family members and the context-dependent roles of proteins like S100A2, which can act as a tumor suppressor in certain tissues, present significant challenges for drug development [4, 14, 16].
