The SARS-CoV-2 spike glycoprotein is a large, trimeric, class I fusion protein that decorates the surface of the virus and mediates viral entry into host cells. The Delta variant (lineage B.1.617.2) features specific mutations in its spike protein—most notably L452R and T478K within the RBD—which increase binding affinity for ACE2 and contribute to higher infectivity. Unique among studied variants, Delta variant spikes promote aggregation of pseudotyped particles through spike-spike tip interactions. The spike glycoprotein is a primary target for neutralizing antibodies elicited by infection or vaccination, making it central both as a therapeutic target and vaccine antigen.
