Overview

SBF2 antisense RNA 1 (SBF2-AS1) is a long non-coding RNA transcribed antisense to the SBF2 gene (myotubularin‐related protein 13; MTMR13). It is approximately 2,700 nucleotides in length and found at chromosome 11p15.4. Predominantly distributed in the cytoplasm, nucleus, and extracellular regions, SBF2-AS1 acts mainly as a competing endogenous RNA, sponging microRNAs such as miR-338-3p and miR-362-3p to regulate gene expression—particularly genes involved in cell cycle, proliferation, and invasion. SBF2-AS1 is frequently upregulated in a range of cancers and is associated with poor prognosis. It interacts with chromatin regulators PRC2 (mainly SUZ12 and EZH2), decreasing tumor suppressor CDKN1A/p21 expression, thereby promoting oncogenesis and tumor growth. Experimental silencing of SBF2-AS1 leads to inhibited proliferation, migration, and invasion in vitro and in vivo, indicating its importance as a potential therapeutic target.

Mechanism of action

Knockdown of SBF2-AS1 inhibits cell proliferation and invasion by sponging tumor suppressor microRNAs such as miR-338-3p, thereby derepressing their targets (e.g., ADAM17, E2F1). SBF2-AS1 binds to PRC2 complex members (SUZ12, EZH2) and guides them to specific gene promoters (e.g., CDKN1A/p21).
