Overview

Schistosoma mansoni calpain (SmCalp) is a calcium-activated neutral cysteine protease primarily localized in the tegumental surface of the Schistosoma mansoni parasite [UniProt, 2024]. It plays a fundamental role in the maintenance and remodeling of the parasite's outer membrane, which is crucial for nutrient uptake and evasion of the host's immune system [PubMed, 2015]. SmCalp is recognized as a significant therapeutic target, with SmCalp1 being a prominent vaccine candidate that has demonstrated protective efficacy in animal models [PubMed, 2018]. Small molecule inhibitors, such as MDL-28170, have been shown to inhibit the proteolytic activity of SmCalp, leading to significant tegumental damage and reduced parasite viability [PubMed, 2014]. The enzyme's accessibility on the parasite surface makes it an attractive target for both immunological and pharmacological interventions [PubMed, 2020]. However, a major challenge in drug development is achieving high selectivity for the schistosome enzyme over human calpain orthologs to avoid potential host toxicity [PubMed, 2020].
