Overview

SEC24 homolog C, COPII coat complex component (SEC24C) is a cytoplasmic adapter protein that is part of the coat protein complex II (COPII), which enables vesicle budding from the endoplasmic reticulum (ER) for transport to the Golgi apparatus[3]. SEC24C belongs to the SEC24 subfamily, with four mammalian paralogs (SEC24A-D), where SEC24C and SEC24D form one subclass based on sequence homology[1]. SEC24C serves as a key cargo adapter, recognizing specific ER export signals and facilitating the selective incorporation of cargo proteins—including certain SNAREs and potentially GPI-anchored proteins—into COPII vesicles for forward trafficking[2][3]. In postmitotic neurons, SEC24C is critical for cell viability and normal brain development; its loss leads to activation of the unfolded protein response and neuronal apoptosis, as well as neural defects such as microcephaly in mice[1]. SEC24C also interacts with viral proteins such as HIV-1 core complexes and Ebola virus VP40, supporting their intracellular transport and contributing to viral pathogenesis[4][5]. No current approved drugs directly target SEC24C, and its primary significance is as a fundamental component of intracellular protein trafficking with essential roles in neural cell biology and viral infection assistance.
