The Secreted frizzled-related protein 1 (SFRP1) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the SFRP1 transcript that controls protein expression levels (Source: NCBI Gene 6422). SFRP1 itself acts as a secreted antagonist of the Wnt signaling pathway by binding to Wnt ligands or Frizzled receptors, thereby inhibiting downstream signaling (Source: UniProt Q8N474). The 3'-UTR contains multiple binding sites for microRNAs (miRNAs), such as miR-27a and miR-454, which typically promote mRNA degradation or inhibit translation (Source: PubMed 23103959, 25600652). In many cancers, including colorectal and breast cancer, SFRP1 is frequently downregulated due to the overexpression of these miRNAs, leading to aberrant Wnt pathway activation and tumor progression (Source: PubMed 28656235). Consequently, the SFRP1 mRNA 3'-UTR is an emerging therapeutic target for RNA-based strategies, such as antisense oligonucleotides or miRNA inhibitors, designed to restore SFRP1 expression and suppress oncogenic signaling. Beyond oncology, the regulation of this region is also implicated in bone density disorders and fibrotic conditions where Wnt signaling plays a key role.
