Overview

The Severe acute respiratory syndrome coronavirus 2 envelope protein (SARS-CoV-2 E protein) is a small, 75-amino acid hydrophobic protein that is essential for the viral life cycle (UniProt P0DTC4). It functions primarily as a viroporin, forming pentameric ion channels in the host's endoplasmic reticulum-Golgi intermediate compartment (ERGIC) membrane, which facilitates viral assembly and budding (PubMed: 32544427). The E protein is also a significant virulence factor; its ion channel activity and interactions with host proteins like PALS1 contribute to the disruption of epithelial barriers and the activation of the NLRP3 inflammasome, leading to severe inflammation (PubMed: 34125054). As a therapeutic target, its ion channel function can be inhibited by small molecules such as amantadine and hexamethylene amiloride (HMA), which may reduce viral titers and mitigate the inflammatory response (PubMed: 33053466). Despite its small size, the E protein's conservation across variants makes it an attractive candidate for broad-spectrum antiviral development.
