Signal transducer and activator of transcription 3 (STAT3) messenger RNA is the nucleic acid template required for the translation of the STAT3 protein, a key mediator of the JAK/STAT signaling pathway (NCBI Gene ID: 6774). This mRNA is a significant therapeutic target because the resulting STAT3 protein acts as a central hub for numerous oncogenic signaling pathways, promoting cell proliferation, survival, and immune suppression in the tumor microenvironment (PubMed: 25934008). While the STAT3 protein itself is often considered "undruggable" by traditional small molecules due to its structural properties, targeting its mRNA transcript using antisense oligonucleotides (ASOs) provides a precise method to silence its expression (Ionis Pharmaceuticals). For instance, Danvatirsen (AZD9150) is a next-generation ASO designed to bind to STAT3 mRNA, triggering its degradation via RNase H and effectively reducing the levels of the oncogenic protein (PubMed: 30530455). This approach is currently being investigated in clinical trials for the treatment of advanced solid tumors and lymphomas, where STAT3 overactivity is a known driver of disease progression and resistance to therapy (ClinicalTrials.gov). Mechanism of action

Antisense oligonucleotides (ASOs) bind to the STAT3 mRNA sequence via complementary base pairing, which triggers RNase H-mediated cleavage of the mRNA, thereby preventing the translation of the STAT3 protein (PubMed: 25934008, Ionis Pharmaceuticals).
