Overview

The Sirtuin 1 (SIRT1) mRNA 3' untranslated region (3' UTR) is a critical regulatory segment of the SIRT1 transcript that governs the post-transcriptional expression of the SIRT1 protein. SIRT1 is a nicotinamide adenine dinucleotide (NAD+)-dependent deacetylase involved in various cellular processes, including DNA repair, metabolism, and longevity (Yamakuchi, 2012, PMID: 22595653). The 3' UTR contains binding sites for numerous microRNAs (miRNAs), such as miR-34a, miR-132, and miR-217, as well as RNA-binding proteins like HuR (ELAVL1), which collectively modulate mRNA stability and translation efficiency (Abdelmohsen et al., 2007, PMID: 17471022). In many age-related and metabolic diseases, SIRT1 expression is downregulated due to the overexpression of inhibitory miRNAs targeting this region (Choi et al., 2013, PMID: 23434671). Consequently, the SIRT1 mRNA 3' UTR has emerged as a therapeutic target for antisense oligonucleotides (ASOs) and miRNA modulators aimed at restoring SIRT1 levels to treat conditions like type 2 diabetes and neurodegeneration. Conversely, in certain cancers where SIRT1 promotes tumor survival, targeting this region with miRNA mimics like MRX34 to decrease SIRT1 expression has been explored, though clinical applications have faced challenges regarding systemic toxicity (Beg et al., 2017, PMID: 28115468).
