Overview

Six-transmembrane epithelial antigen of the prostate 1 (STEAP1) is a cell surface protein that is highly overexpressed in several types of cancer, most notably prostate cancer and Ewing sarcoma, while showing limited expression in normal tissues. It belongs to the STEAP family of metalloreductases and is characterized by a structure containing six transmembrane domains. Biologically, STEAP1 functions as a metalloreductase, facilitating the reduction of iron and copper, which supports cellular metabolism and promotes tumor cell proliferation and survival by modulating oxidative stress and signaling pathways. Due to its high tumor specificity and accessible extracellular loops, STEAP1 has emerged as a promising therapeutic target for advanced malignancies. Current drug development efforts focus on utilizing STEAP1 as a target for antibody-drug conjugates (ADCs) and bispecific T-cell engagers (BiTEs), such as xalurmabig (AMG 509), which aim to selectively eliminate cancer cells by delivering cytotoxic payloads or redirecting immune effector cells.
