Overview

The **smear layer** in root canals is a thin layer (2–5 micrometers) of microcrystalline and organic debris created on the canal wall after root canal instrumentation[1][2][4][5]. It contains residual organic tissue, pulp remnants, dentin debris, bacteria, bacterial byproducts, necrotic material, and can be heterogeneous in composition[1][2][4][5]. The layer may occlude dentinal tubules and affect both the disinfection of the root canal system and the sealing of canal filling materials[2][5]. Chemomechanical techniques, especially with irrigants like sodium hypochlorite and EDTA, are used to dissolve or remove the smear layer as an essential part of endodontic therapy, although no method achieves complete removal in all cases[5][6]. The presence or absence of the smear layer can influence endodontic outcomes, bacterial penetration, and microleakage, but it is not itself considered a therapeutic target in the sense of a discrete protein or molecular structure suitable for drug targeting[1][2][5][6].
