Overview

Smooth muscle lineage cells (SMCs) are specialized, involuntary muscle cells found within the walls of various internal organs, including blood vessels, the respiratory tract, and the gastrointestinal system (StatPearls, 2023). Their fundamental biological role involves the regulation of luminal diameter and organ tone through coordinated contraction and relaxation, which is critical for maintaining hemodynamic stability and facilitating the movement of contents through hollow organs (Nature Reviews Cardiology, 2020). Unlike skeletal or cardiac muscle, SMCs maintain a high degree of phenotypic plasticity, allowing them to transition between a contractile state and a synthetic, proliferative state in response to environmental cues or vascular injury (Circulation Research, 2015). This plasticity is a double-edged sword; while essential for wound healing, it is a primary driver of pathological remodeling in diseases such as atherosclerosis, hypertension, and asthma (Journal of Clinical Investigation, 2018). Although "Smooth muscle lineage cells" refers to a broad cell population rather than a single molecular entity, they are the primary site of action for several major drug classes, including calcium channel blockers, nitrates, and endothelin receptor antagonists (PubMed, 2021).
