Overview

Smoothened (SMO) is a Class F GPCR and a key component of the Hedgehog signaling pathway, essential for development and tissue homeostasis. It is inhibited by Patched (PTCH1) in the absence of Hedgehog ligands and activated upon ligand binding to PTCH1. Aberrant SMO activation is implicated in several cancers, particularly basal cell carcinoma and medulloblastoma, making it a therapeutic target. However, resistance to SMO inhibitors is a growing concern.

Mechanism of action

Upon Hedgehog ligand binding to PTCH1, SMO is activated and interacts with Gi proteins to propagate downstream signaling, regulating gene expression.
