Overview

Sodium-hydrogen exchanger 1 (NHE1), encoded by the SLC9A1 gene, is a ubiquitous plasma membrane transporter that plays a pivotal role in maintaining intracellular pH and cell volume by exchanging one intracellular proton for one extracellular sodium ion [1, 2]. The NHE1 mRNA transcript is a critical regulatory point for the expression of this transporter, which is often upregulated in pathological conditions such as heart failure and various cancers [3, 4]. In cardiovascular disease, overactivity of the NHE1 protein leads to sodium and calcium overload, contributing to ischemia-reperfusion injury and cardiac remodeling [4]. In oncology, high levels of NHE1 mRNA and protein facilitate an acidic tumor microenvironment, which promotes basement membrane degradation, tumor invasion, and metastasis [3, 5]. Therapeutic strategies targeting NHE1 include small-molecule inhibitors that bind to the protein and emerging RNA-based therapies, such as siRNAs or antisense oligonucleotides, designed to degrade NHE1 mRNA and reduce its translation [5, 6].
