Sperm associated antigen 9 (SPAG9) is a scaffold protein belonging to the JNK-interacting protein (JIP) family, primarily involved in the regulation of the c-Jun N-terminal kinase (JNK) signaling pathway (UniProt: Q5T210). It is characterized as a cancer-testis antigen (CTA), meaning its expression is typically restricted to the immune-privileged testis but becomes highly expressed in various human cancers, such as breast, cervical, and lung carcinomas (PubMed: 21533181). In malignant cells, SPAG9 promotes tumor growth, migration, and invasion, making it a significant driver of oncogenesis (PubMed: 25600155). Because of its restricted expression pattern and vital role in cancer progression, it serves as an ideal target for immunotherapy. Therapeutic strategies, such as dendritic cell-based vaccines or T-cell therapies, aim to elicit a robust immune response involving antigen-specific T cells and natural killer (NK) cells to selectively destroy SPAG9-positive tumor cells (PubMed: 17460773). This approach leverages the high immunogenicity of SPAG9 to induce both cellular and humoral immune responses against the tumor.
