Sperm protein associated with the nucleus, X chromosome, family member A2 (SPANXA2) is a member of the SPANX family of cancer/testis antigens that encode testis-specific proteins predominantly localized to unique regions within the sperm nucleus[2][7]. The gene is part of a cluster on the X chromosome, and its protein product typically localizes to nuclear vacuoles and the redundant nuclear envelope, suggesting a biochemical role in the structural organization of spermatozoa nuclei as well as a marker for studying sperm-specific subnuclear compartments[2][7]. SPANXA2, like other SPANX antigens, is expressed mainly in the testis under healthy conditions but is also aberrantly expressed in certain cancers, such as melanoma, bladder carcinoma, and myeloma, where it is considered a cancer/testis antigen[7]. These features make SPANXA2 a potential immunotherapy target and a relevant biomarker for both reproductive biology and oncological research[7]. No approved therapeutic drugs are currently known to act directly on this protein, but it is under study as a vaccine antigen in oncology[7][2].
